延胡索配方顆粒多指標成分的近紅外光譜快速檢測方法

[摘要]目的建立延胡索配方顆粒水分、醇溶性浸出物、延胡索乙素、去氫延胡索甲素多指標的近紅外光譜快速檢測方法。方法以已知的延胡索配方顆粒水分、醇溶性浸出物、延胡索乙素、去氫延胡索甲素含量值為對照,運用偏最小二乘法(PLS)選擇合適的光譜范圍及光譜預處理方法建立近紅外定量模型,并對驗證集樣品各指標成分的含量進行驗證。結果水分、醇溶性浸出物、延胡索乙素、去氫延胡索甲素預測值與實測值的平均相對偏差分別為1.18%、2.19%、3.14%、1.73%,預測相關系數 R2分別為0.979、0.915、0.948、0.962,準確度較高。結論研究建立的近紅外定量模型可應用于延胡索配方顆粒中水分、醇溶性浸出物、延胡索乙素、去氫延胡索甲素含量4個指標成分的快速檢測,并可為其他中藥配方顆粒的快速檢測提供參考。[關鍵詞]近紅外光譜;延胡索;配方顆粒;快速檢測

A rapid determination method of multiple index components in rhizoma corydalis dispensing granules by near -infrared spectroscopy

[Abstract] Objective To establish a rapid determination method of moisture, alcohol-soluble extract, tetrahydropalmatine and dehydrocorydaline in rhizoma corydalis dispensing granules by near-infrared (NIR) spectroscopy. Methods A quantitative NIR model was established by using appropriate spectral range and spectral pre-processing method selected with the partial least squares (PLS) method, taking the contents of known moisture, alcohol-soluble extract, tetrahydropalmatine and dehydrocorydaline in rhizoma corydalis dispensing granules as the reference. Results The relative mean deviations of the predicted and measured values of moisture, alcohol- soluble extract, tetrahydropalmatine and dehydrocorydaline were 1.18%, 2.19%, 3.14% and 1.73%, respectively, and the predicted correlation coefficients (R2) were 0.979, 0.915, 0.948 and 0.962, respectively, with high accuracy. Conclusion The established NIR quantitative model can be applied to the rapid determination of the contents of moisture, alcohol-soluble extract, tetrahydropalmatine and dehydrocorydaline in rhizoma corydalis dispensing granules, and can provide a reference for the rapid determination of other traditional Chinese medicine dispensing granules.

延胡索配方w粒是以中藥飲片延胡索為原料,運用現代制藥工藝,制備成供中醫臨床上可隨機組方、即沖即飲的顆粒劑型。醋延胡索配方顆粒國家藥品標準[1]中的檢測過程耗時耗力,無法滿足生產企業即時、快速分析的需要。
近紅外光譜技術是一種利用近紅外光譜區包含的有效信息,依靠計算機技術對物質進行快速定性或定量的分析技術,具有高效便捷、無破壞性、無污染、結果準確的優勢[2],在藥物分析領域的應用最早出現在20世紀60年代晚期[3],在中國、美國、歐洲、日本藥典中均有收錄[4]。目前該技術已逐步應用于中藥行業[5-6],如品種的真偽鑒別[7-8]、不同產地藥材鑒別[9-10]、不同生產廠家鑒別[11]、炮制程度[12]、有效成分含量快速測定[13-15]等。
